Выбор метода выделения фракции ядросодержащих клеток из пуповинной крови как основа организации банка пуповинной крови
Аннотация
При организации деятельности банка пуповинной крови актуальной является сравнительная оценка различных технических приемов обработки пуповинной крови для обеспечения максимально высокого выхода клеток с учетом соотношения «затраты - эффективность». Проведено сравнительное исследование эффективности выделения фракции ядросодержащих клеток из пуповинной крови методами двойного центрифугирования, полуавтоматической и автоматической сепарации. Полуавтоматическая и автоматическая системы сепарации эффективнее метода двойного центрифугирования по основным качественным показателям клеточного концентрата: общему количеству ядросодержащих клеток, выходу ядросодержащих клеток. Показано, что полуавтоматическая система выделения фракции ядросодержащих клеток требует наименьшего количества времени для обработки одного образца. На основании полученных данных такая система предложена как оптимальная при большом (более 5) ежедневном поступлении образцов пуповинной крови в банк пуповинной крови для обработки и хранения.

Isolation of nucleated cells fraction from umbilical cord blood -choice of method
Abstract
A comparative study of the effectiveness of nucleated cells fraction isolation from umbilical cord blood. The methods of double centrifugation, automatic and semi- automatic separation was compared. It was shown that semi-automatic and automatic processing methods were more efficient then the double centrifugation by cell concentrate key quality indicators: the total number of nucleated cells, nucleated cells recovery. Also, the semi-automatic nucleated cells fraction isolation method requires the least amount of time for processing a single sample. Based on these data such a system has been proposed as optimal for large (more than 5) daily admission of cord blood samples into cord blood bank.
