Изучение интенсивности высвобождения аминексила из мазевых основ

Аннотация

Облысение доставляет людям значительный психологический дискомфорт, представляя тем самым значительную медико-социальную проблему. Миноксидил вот уже на протяжении более двух десятков лет остается практически единственным препаратом, с уверенностью рекомендованным дерматологами в качестве средства для восстановления роста волос. Однако для получения терапевтического эффекта необходимо его длительное применение. Кроме того, ограничением на использование миноксидила является наличие у пациентов нарушений барьерной функции кожи (псориаз, экзема), ввиду данных о возможности возникновения аллергического и контактного дерматита после наружного применения миноксидила, особенно в высоких концентрациях. В связи с этим представляется перспективным использование производных миноксидила, обладающих лучшей переносимостью, в частности аминексила (копексила) - 2,4-диамино- пиримидин-3-N-oксида. В настоящее время на фармацевтическом рынке Украины аминексил представлен в виде космецевтических препаратов (спреи и растворы для втираний в волосистую часть кожи головы, относящихся к высокоценовому сегменту и поэтому малодоступных для широкого использования. Разработка на основе комбинации аминексила и жирного масла семян арганы колючей (Argania spinosa) мягкой лекарственной формы или парафармацевтического средства в форме мази (крема, геля) для наружного применения, обеспечивающего укрепление, восстановление волос, предотвращающего их выпадение, устраняющего сухость кожи головы с использованием доступных отечественных технологий и соответствующего оборудования представляет собой научный и практический интерес для отечественной дерматологии, позволит предложить реальную альтернативу дорогостоящим зарубежным препаратам. Целью настоящей работы является биофармацевтическое обоснование рационального состава мягкой лекарственной формы аминексила в комбинации с аргановым маслом для наружного применения. Высвобождение аминексила из мазевых композиций изучали методом равновесного диализа по Крувчинскому. В качестве диализной среды с учетом растворимости аминексила использовали 0,1 М раствора хлористоводородной кислоты. Концентрацию высвободившегося через 30 минут вещества определяли спектрофотометрически. Установлено, что вид основы-носителя оказывает значимое влияние на высвобождение аминексила из мазей для наружного применения. Дисперсионный анализ результатов исследований показал, что оптимальную высвобождаемость аминексила из мазей обеспечивает гидрофильный носитель на основе натрий-карбоксиметилцеллюлозного глицерогеля.

Abstract

Alopecia causes a significant psychological discomfort for people and therefore represents an important medical and social problem. During more then two tens years minoxidil has been practically the only drug which is recommended by the dermatologists as a medication for the recovery of hair growth. But the long use is needed for the therapeutic effect. Besides the skin barrier dysfunction (psoriasis, eczema) in humans according to data about possibility of allergic and contact dermatitis after external administration of minoxidil especially in high concentration limits the use.In this connection the use of minoxidil derivates with better tolerance, in particular, aminexil (kopexil)- 2,4-diaminopirimidin-3-N-oxydeisperspective. Atpresentonthe Ukrainianpharmaceutical market aminexil is presented by the cosmeceuticals (sprays and solutions for rubbing in skin of scalp) which belong to high concentrated segment and so are the inaccessible for the wide use.Development on the base of aminexil and argan Argania spinosa oil combination the semisolid dosage form or parapharmaceuticals in form of ointment (cream, gel) for external use providing with hair strengthening and recovery, preventing its shedding, removing the head skin dryness with using the accessible domestic technologies and appropriate devices is of scientific and practical interest to domestic dermatology, will allow proposing the real alternative to expensive imported medications.The aim of this work is biopharmaceutical substantiation of rational composition of aminexil semisolid dosage form in combination with argan oil for external use.Aminexil releasing from ointment compositions was studied by Kruvchinsky equilibrium dialysis method. As a dialysis medium according aminexil solubility the 0,1 M hydrochloric acid was used. Concentration of released substance after 30 min was determined spectrophotometry.In was established that the sort of base has a significant influence on the aminexil releasing from the ointments for external use. Variance analysis of investigation’s results presented that the optimal aminexil releasing from ointment is provided with the hydrophilic device on the base of sodium carboxymethylcellulose glycerogel.
