Значение лабораторной информационной системы в контроле процессинга гаплоидентичной трансплантации гемопоэтических стволовых клеток с деплецией TCRaß/CD19

Аннотация

В материале представлены исторические аспекты разработки и оптимизации гаплоидентичной трансплантации гемопоэтических стволовых клеток, а также ее преимущества на современном этапе при проведении аллогенной трансплантации. Рассмотрена технология проведения лабораторного анализа методом проточной цитометрии деплеции TCRaß-клеток и CD19, включая описание методики до и после иммуномагнитного разделения на аппарате CliniMACSPlusSystem. Приведена методология применения протокола процессов сбора и анализа данных деплеции TCRaß/CD19 цитометрического исследования и его интеграции и организации с лабораторной информационной системой в лаборатории трансплантационной иммунологии и иммунотерапии гемобластозов ФГБУ «ФНКЦ ДГОИ им. Дмитрия Рогачева» Минздрава России. Показаны результаты анализа лабораторно-диагностических данных, включая оценку качества деплеции TCRaß/CD19.

Abstract

Historical aspects of the development and optimization of the haploidentical hematopoietic stem cells transplantation and its advantages in the modern era are presented in the article. The TCRaß/CD19 depletion laboratory analysis technology using flow cytometry including a discription of the method before and after immuno-magnetic separation on the unit CliniMACSPlusSystem is shown. Methodology of application of the flow cytometric data collection and analysis protocol of the TCRaß/CD19 depletion and its integration with the laboratory information system in Laboratory of the Transplant Immunology and Immunotherapy of Hemoblastosis, Federal Scientific Clinical Centre of Pediatric Hematology Oncology Immunology named after Dmitry Rogachev Ministry of Health of the Russian Federation are presented. The results of laboratory data analysis including TCRaß/CD19 depletion quality assessment are also shown.
